RESUMO

Urbanization has altered land surface properties driving changes in micro-climates. Urban form influences people's activities, environmental exposures, and health. Developing detailed and unified longitudinal measures of urban form is essential to quantify these relationships. Local Climate Zones [LCZ] are a culturally-neutral urban form classification scheme. To date, longitudinal LCZ maps at large scales (i.e., national, continental, or global) are not available. We developed an approach to map LCZs for the continental US from 1986 to 2020 at 100 m spatial resolution. We developed lightweight contextual random forest models using a hybrid model development pipeline that leveraged crowdsourced and expert labeling and cloud-enabled modeling - an approach that could be generalized to other countries and continents. Our model achieved good performance: 0.76 overall accuracy (0.55-0.96 class-wise F1 scores). To our knowledge, this is the first high-resolution, longitudinal LCZ map for the continental US. Our work may be useful for a variety of fields including earth system science, urban planning, and public health.

AIMS: Pancreatic islets undergo critical development and functional maturation during the perinatal period when they are highly sensitive to microenvironment. We aim to determine the effects and mechanisms of gestational diabetes mellitus (GDM) hypermetabolic stress on glucose homeostasis in pregnant mice and functional maturation of the islets of their offspring. MAIN METHODS: Exosomes were extracted from the umbilical vein blood of individuals with or without GDM for administration to pregnant mice. The blood glucose, serum insulin, glycosylated hemoglobin, and lipopolysaccharide levels were measured in pregnant mice. The expression and localization of insulin, glucagon, PC1/3, PDX1, and p-S6 in the islets of neonatal rats were continuously monitored using immunofluorescence to evaluate their functional status. Primary islet cells were cultured and treated with GDM exosomes and exendin to determine the expression of GLP-1R, AKT, p-AKT, and p-S6 via western blotting. KEY FINDINGS: GDM exosomes induced remarkable oral glucose intolerance, hyperinsulinemia, and metabolic inflammation in pregnant mice. The islets of GDM offspring exhibited high insulin, glucagon, PC1/3, PDX1, and p-S6 expression at and after birth, and activation of the local GLP-1/GLP-1R axis. The functional maturation of normal-offspring islets did not commence until after birth, while it was activated prior to birth in GDM offspring, seriously disrupting the whole process. GDM exosomes activated the GLP-1/GLP-1R axis between α and ß cells, and stimulated functional maturation of ß cells via the Akt-mTORC1-pS6 pathway. SIGNIFICANCE: These findings provide preliminary insights into the mechanisms underlying the high incidence of diabetes in the offspring of mothers with GDM.

SECRETORY13 (SEC13) is an essential member of the coat protein complex II (COPII), which was reported to mediate vesicular-specific transport from the endoplasmic reticulum (ER) to the Golgi apparatus and plays a crucial role in early secretory pathways. In Arabidopsis, there are two homologous proteins of SEC13: SEC13A and SEC13B. SUPPRESSOR OF FRIGIDA 4 (SUF4) encodes a C2H2-type zinc finger protein that inhibits flowering by transcriptionally activating the FLOWERING LOCUS C (FLC) through the FRIGIDA (FRI) pathway in Arabidopsis. However, it remains unclear whether SEC13 proteins are involved in Arabidopsis flowering. In this study, we first identified that the sec13b mutant exhibited early flowering under both long-day and short-day conditions. Quantitative real-time PCR (qRT-PCR) analysis showed that both SEC13A and SEC13B were expressed in all the checked tissues, and transient expression assays indicated that SEC13A and SEC13B were localized not only in the ER but also in the nucleus. Then, we identified that SEC13A and SEC13B could interact with SUF4 in vitro and in vivo. Interestingly, both sec13b and suf4 single mutants flowered earlier than the wild type (Col-0), whereas the sec13b suf4 double mutant flowered even earlier than all the others. In addition, the expression of flowering inhibitor FLC was down-regulated, and the expressions of flowering activator FLOWERING LOCUS T (FT), CONSTANS (CO), and SUPPRESSOR OF OVEREXPRESSION OF CO 1 (SOC1) were up-regulated in sec13b, suf4, and sec13b suf4 mutants, compared with Col-0. Taken together, our results indicated that SEC13B interacted with SUF4, and they may co-regulate the same genes in flowering-regulation pathways. These results also suggested that the COPII component could function in flowering in Arabidopsis.

Plants deploy receptor-like kinases and nucleotide-binding leucine-rich repeat receptors to confer host plant resistance (HPR) to herbivores1. These gene-for-gene interactions between insects and their hosts have been proposed for more than 50 years2. However, the molecular and cellular mechanisms that underlie HPR have been elusive, as the identity and sensing mechanisms of insect avirulence effectors have remained unknown. Here we identify an insect salivary protein perceived by a plant immune receptor. The BPH14-interacting salivary protein (BISP) from the brown planthopper (Nilaparvata lugens Stål) is secreted into rice (Oryza sativa) during feeding. In susceptible plants, BISP targets O. satvia RLCK185 (OsRLCK185; hereafter Os is used to denote O. satvia-related proteins or genes) to suppress basal defences. In resistant plants, the nucleotide-binding leucine-rich repeat receptor BPH14 directly binds BISP to activate HPR. Constitutive activation of Bph14-mediated immunity is detrimental to plant growth and productivity. The fine-tuning of Bph14-mediated HPR is achieved through direct binding of BISP and BPH14 to the selective autophagy cargo receptor OsNBR1, which delivers BISP to OsATG8 for degradation. Autophagy therefore controls BISP levels. In Bph14 plants, autophagy restores cellular homeostasis by downregulating HPR when feeding by brown planthoppers ceases. We identify an insect saliva protein sensed by a plant immune receptor and discover a three-way interaction system that offers opportunities for developing high-yield, insect-resistant crops.

More than 11 million Americans reside within 150 meters of a highway, an area of high air pollution exposure. Traffic congestion further contributes to environmental pollution (e.g., air and noise), but its unique importance for population health is unclear. We hypothesized that degraded environmental quality specifically from traffic congestion has harmful impacts on fetal growth. Using a population-based cohort of births in Texas (2015-2016), we leveraged connected vehicle data to calculate traffic congestion metrics around each maternal address at delivery. Among 579,122 births, we found consistent adverse associations between traffic congestion and reduced term birth weight (8.9 grams), even after accounting for sociodemographic characteristics, typical traffic volume, and diverse environmental coexposures. We estimated that up to 1.2 million pregnancies annually may be exposed to traffic congestion (27% of births in the United States), with ~256,000 in the highest congestion zones. Therefore, improvements to traffic congestion may yield positive cobenefits for infant health.

Lignocellulose, which contains cellulose, hemicellulose and lignin, is one of the most important factors determining the rate and quality of compost decomposition, and the microbial community composition affects the rate of lignocellulose decomposition. Interactions between microbial taxa contribute significantly to ecosystem energy flow and material cycling. However, it is not clear how interactions between microbial taxa affect the degradation of lignocellulose during the composting process. For this reason we carried out aerobic co-composting experiments with maize straw and cattle manure to explore the contribution of microbial community diversity and the interaction between taxa to lignocellulosic degradation. The results showed that moisture and temperature had the greatest effect on microbial communities during composting and that lignocellulose degradation was dominated by microbial co-occurrence networks rather than microbial community diversity. Overall co-occurrence network and bacterial-fungal interactions explained 23.9-84.1 % of lignocellulosic degradation, whereas microbial diversity only accounted for 24.6-31.5 %. Interestingly, keystone taxa analysis of the microbial co-occurrence networks revealed that low-abundance taxa influenced microbial interactions driving lignocellulose degradation. Our results provide a new perspective for understanding lignocellulose degradation during composting, offering insights into important microbial interaction mechanisms for improving compost quality and efficiency.

In order to verify the technology of the membrane diffractive imaging system for Chinese next generation geo-stationary earth orbit (GEO) satellite, a series of ground experiments have been carried out using a membrane optical camera with 80 mm aperture (Φ80) lens. The inherent chromatic aberration due to diffractive imaging appears in the obtained data. To address the issue, an effective color restoration algorithm framework by matching, tailoring, and non-linearly stretching the image histograms is proposed in this letter. Experimental results show the proposed approach has good performances in color restoration of the diffractive optical images than previous methods. The effectiveness and robustness of the algorithm are also quantitatively assessed using various color deviation indexes. The results indicate that the chromatic aberration of diffractive images can be effectively removed by about 85%. Also, the proposed method presents reasonable computational efficiency.

The brown planthopper (BPH), Nilaparvata lugens (Stål), is a phloem sap-feeding insect. During feeding on rice plants, BPH secretes salivary proteins with potential effector functions, which may play a critical role in the plant-insect interactions. However, a limited number of BPH effector proteins have been identified to date. Here, we sequenced the salivary gland transcriptomes of five BPH populations and subsequently established a N. lugens secretome consisting of 1,140 protein-encoding genes. Secretome analysis revealed the presence of both conserved and rapidly evolving salivary proteins. A screen for potential effectors that elicit responses in the plant was performed via the transient expression analysis of 64 BPH salivary proteins in Nicotiana benthamiana leaves and rice protoplasts. The salivary proteins Nl12, Nl16, Nl28, and Nl43 induced cell death, whereas Nl40 induced chlorosis and Nl32 induced a dwarf phenotype in N. benthamiana, indicating effector properties of these proteins. Ectopic expression of the six salivary proteins in N. benthamiana upregulated expression of defense-related genes and callose deposition. Tissue expression analysis showed a higher expression level of the six candidate effectors in salivary glands than in other tissues. Subcellular localization and analysis of the domain required for cell death showed a diverse structure of the six effectors. Nl28, Nl40, and Nl43 are N. lugens specific; in contrast, Nl12, Nl16, and Nl32 are conserved among insects. The Nl40 family has numerous isoforms produced by alternative splicing, exemplifying rapid evolution and expansion of effector proteins in the BPH. Our results suggest a potential large effector repertoire in BPH and a higher level of effector conservation exist in BPH compared with that in plant pathogens.

Most of the planet's population currently lives in urban areas, and urban land expansion is one of the most dramatic forms of land conversion. Understanding how cities evolve temporally, spatially, and organizationally in a rapidly urbanizing world is critical for sustainable development. However, few studies have examined the coevolution of urban attributes in time and space simultaneously and the adequacy of power law scaling across cities and through time, particularly in countries that have experienced abrupt, widespread, political and economic changes. Here, we show the temporal coevolution of multiple physical, demographic, socioeconomic, and environmental attributes in individual cities, and the cross-city scaling of urban attributes at six time points (i.e., 1978, 1990, 1995, 2000, 2005, and 2010) in 32 major Chinese cities. We found that power law scaling could adequately characterize both the cross-city scaling of urban attributes across cities and the longitudinal scaling describing the temporal coevolution of urban attributes within individual cities. The cross-city scaling properties demonstrated substantial changes over time signifying evolved social and economic forces. A key finding was that the cross-city linear or superlinear scaling of urban area with population contradicts the theoretical sublinear power law scaling proposed between infrastructure and population. Furthermore, the cross-city scaling between area and population transitioned from linear to superlinear over time, and the superlinear scaling in recent times suggests decreased infrastructure efficiency. Our results demonstrate a diseconomy of scale in urban areal expansion that indicates a significant waste of land resources in the urbanization process. Future planning efforts should focus on policies that increase urban land use efficiency before continuing expansion.

Based on ozone observation data from urban stations and the Dingling (DL) background station, we investigated the trend of ozone concentrations in Beijing during 2004-2015. For urban stations, both O3_1 h and O3_8 h increased stably with a clear and significant linear pattern and the increase rate was notably higher during the period of May to Sep. Meanwhile, the variation of O3_1 h and O3_8 h for the DL station did not demonstrate a regular pattern. During this period, the differences between the diurnal peak of ozone concentrations at the DL background station and urban stations decreased significantly due to the rapid urbanization of Beijing. Furthermore, we examined simultaneous variations of ozone and its precursors during 2015 Grand Military Parade and 2014 APEC meeting and evaluated the performances of different emission-reduction measures during the two specific events. For 2015 Grand Military Parade, emission-reduction measures were implemented 14 days in advance, which led to a notable decrease of ozone concentrations during the Parade period. For 2014 APEC meeting, emission-reduction measures were not implemented in advance, which led to incomplete VOCs reduction and high VOCs/NOx values, and thus a significant increase of ozone concentrations during the APEC period. The emission-reduction measures during APEC and PARADE periods both slowed down the accumulation and cut down the concentration peaks of ozone. We also analyzed simultaneous concentration variations of ozone and its precursors in long time-series. The results proved that compared with other precursors, NO2/NO was an effective indicator for ozone concentration in Beijing, especially in urban areas. The findings from this research provide useful reference for better monitoring and managing ozone concentrations in Beijing and other cities through properly designed and implemented emission-reduction measures.

The brown planthopper (BPH) and white-backed planthopper (WBPH) are the most destructive insect pests of rice, and they pose serious threats to rice production throughout Asia. Thus, there are urgent needs to identify resistance-conferring genes and to breed planthopper-resistant rice varieties. Here we report the map-based cloning and functional analysis of Bph6, a gene that confers resistance to planthoppers in rice. Bph6 encodes a previously uncharacterized protein that localizes to exocysts and interacts with the exocyst subunit OsEXO70E1. Bph6 expression increases exocytosis and participates in cell wall maintenance and reinforcement. A coordinated cytokinin, salicylic acid and jasmonic acid signaling pathway is activated in Bph6-carrying plants, which display broad resistance to all tested BPH biotypes and to WBPH without sacrificing yield, as these plants were found to maintain a high level of performance in a field that was heavily infested with BPH. Our results suggest that a superior resistance gene that evolved long ago in a region where planthoppers are found year round could be very valuable for controlling agricultural insect pests.

BROWN PLANTHOPPER RESISTANCE14 (BPH14), the first planthopper resistance gene isolated via map-based cloning in rice (Oryza sativa), encodes a coiled-coil, nucleotide binding site, leucine-rich repeat (CC-NB-LRR) protein. Several planthopper and aphid resistance genes encoding proteins with similar structures have recently been identified. Here, we analyzed the functions of the domains of BPH14 to identify molecular mechanisms underpinning BPH14-mediated planthopper resistance. The CC or NB domains alone or in combination (CC-NB [CN]) conferred a similar level of brown planthopper resistance to that of full-length (FL) BPH14. Both domains activated the salicylic acid signaling pathway and defense gene expression. In rice protoplasts and Nicotiana benthamiana leaves, these domains increased reactive oxygen species levels without triggering cell death. Additionally, the resistance domains and FL BPH14 protein formed homocomplexes that interacted with transcription factors WRKY46 and WRKY72. In rice protoplasts, the expression of FL BPH14 or its CC, NB, and CN domains increased the accumulation of WRKY46 and WRKY72 as well as WRKY46- and WRKY72-dependent transactivation activity. WRKY46 and WRKY72 bind to the promoters of the receptor-like cytoplasmic kinase gene RLCK281 and the callose synthase gene LOC_Os01g67364.1, whose transactivation activity is dependent on WRKY46 or WRKY72. These findings shed light on this important insect resistance mechanism.

The concentrations of Cr, Co, Ni, Cu, Zn, Pb, Cd, As and Hg in surface sediment samples of Yangtze River collected in 2007 were analyzed and evaluated. The results indicated that the concentrations of Cu, Zn, Pb, Cd, As and Hg were significantly higher than those measured in 1990s. Principal component analysis showed that the cumulative proportion of the first three components accounted for 86.75% of the total variable, indicating the three major sources of heavy metals were industrial and mining wastewater, weathering and erosion of rocks, and urban electroplating industry wastewater and natural sources. Geoaccumulation index (Igeo) and enrichment factors (EF) also showed that the surface sediments of the Yangtze River were not contaminated with Cr, Co and Ni, lightly contaminated with Cu, Zn, As and Hg, and majorly contaminated with Ph and Cd. The ecological hazards for the heavy metals in the sediments were evaluated with the Hakanson ecological risk index. It was concluded the ecological hazards for each metal in a descending order were Cd > Hg > As > Zn > Pb > Cu > Co > Ni > Cr. The comprehensive index of potential ecological risks for metals indicated that 36% of the 61 sites had moderate potential ecological risks. Three sites had a high potential ecological risk, namely, Chongqing site of the main Yangtze River, Zishui Dongting Lake and Xinjiang site, whereas Xiangjiang Hengyang section, Xiangjiang Zhuzhou section, Xiangjiang Dongting Lake entrance, Dongting Lake and Shunan River belonged to the areas with extremely high potential ecological risk.

In order to analyze and evaluate different trace metals on surface water of the Changjiang River, concentrations of dissolved trace metals (Cu, Ni, Fe, Co, Sc, Al, Zn, Pb, Cd, Se, As, Cr, and Hg), major elements (Ca and Mg), and nutrient (NO3- were measured. Samples were taken at 76 positions along Changjiang River in flood and dry seasons during 2007-2008. Spatial distributions identified two main large zones mainly influenced by mineral erosion (sites 1-22) and anthropogenic action (sites 23-76), respectively. Principal component analysis (PCA) and hierarchical cluster analysis were used to identify the variance distinguishing the origin of water. Four significant components were extracted by PCA, explaining 74.91% of total variable. Cu, Ni, Fe, Co, Sc, Al, Ca, and Mg were mainly associated with the weathering and erosion of various rocks and minerals, while an anthropogenic source was identified for Cd and As. Although erosion was one source of Pb and Zn, they were also input by atmospheric deposition and industrial pollutions. NO3- and Se were mainly associated with agriculture activities. However, Hg and Cr showed different sources. CA confirmed and completed the results obtained by PCA, classifying the data into two large groups representing different areas. Group 1 referred to the upper reaches which represented samples mainly corresponding to natural background areas. Group 2 referred to the middle and lower reaches including samples under anthropogenic influence. Meanwhile, group 2 was subdivided into three new groups, representing agricultural, industrial, and various artificial pollution sources, respectively.
